Reduced levels of F2RL3 methylation in whole blood in thrombotic diseases

  • 目的:探讨血栓形成性疾病患者F2R样凝血酶/胰蛋白酶受体3(F2RL3)基因甲基化水平,以评价F2RL3基因甲基化鉴别血栓性疾病的效能。方法:收集246例血栓性患者的EDTA-K2抗凝全血标本,包括颈内静脉血栓(IJVT)37例、下腔静脉栓塞(IVCT)49例、下肢静脉栓塞(LEVT)32例、心肌梗死(MI)52例、脑梗死(CI)76例和健康对照组(Ctrl)76例,采用实时荧光定量PCR技术检测全血F2RL3基因启动子甲基化水平,并分析其与危险因素的相关性。结果:①246例血栓性疾病患者全血F2RL3基因甲基化水平为(0.49±0.11),低于Ctrl组(0.60±0.15),差异有统计学意义(P<0.05)。②静脉血栓患者(VT组)全血F2RL3基因甲基化水平为(0.50±0.08),动脉血栓者(AT组)F2RL3基因甲基化水平为(0.49±0.12),均低于Ctrl组(P<0.05)。③IVCT组患者(0.48±0.08)、LEVT组患者(0.49±0.11)和CI患者(0.48±0.09)F2RL3基因甲基化水平低于Ctrl组,差异有统计学意义(P<0.05)。结论:全血F2RL3基因甲基化水平降低可辅助诊断血栓形成性疾病。
