Preliminary study on relationship between A blood subtype and glycosyl transferase activity

摘要: 目的: 通过研究A亚型等位基因及其编码糖基转移酶的活性,进而了解不同A亚型的抗原性。方法: 血型表型的定型采用微孔板法初检,试管法确认;糖基转移酶活性的检测采用血清学试管法;A血型亚型等位基因的检测采用DNA分子测序的方法。结果: 在与抗A1不凝集的8例A亚型、24例A亚型B的标本中,无α-1、3-N-乙酰半乳糖基转移酶活性的分别为7例和24例;在与抗A1弱凝集的6例A型、13例AB型标本中,出现酶活性的分别为1例和4例。95.45%(42/44)的A亚型无α-1、3-N-乙酰半乳糖基转移酶活性。结论: 从本次初步研究看绝大部分的A亚型无α-1、3-N-乙酰半乳糖基转移酶活性。不同A等位基因其A亚型的抗原性有差别。

Abstract: Objective: To investigate the different antigenicity of A subtype by analysing the A subtype alleles and activity of its glycosyl transferase.Method: A total of 10 012 specimens were tested by microtiter plates for ABO, then abnormal specimens were scanned and measured by tube test. Exon 6 and exon 7 of ABO gene were analyzed by PCR and sequence analysis. Result: In no agglutination of anti-A1 of 8cases of subtype A and 24 cases of speimens of AsubtypeB, no α-1,3-N-acetyl galactose glycosyl transferase activity were 7 cases and 24 cases respectively. In weak agglutination of A subtype and AsubtypeB specimens, the enzyme activity cases were 1/6 and 4/13. 95.45%(42/44) speimens of A subtype were lack of glycosyl transferase activity. Conclusion: The most part of A subtype of α-1,3-N-acetyl galactose glycosyl transferase activity were no detected. And antigenicity of A subtype may have difference among different A alleles.-
