Knops blood group polymorphisms of blood donors among Han populations in Jiangsu area

摘要: 目的: Knops血型系统的抗原由Ⅰ型补体受体(CR1)基因编码,通过调查献血者CR1基因7个单核苷酸多态性,了解江苏地区Knops血型系统抗原分布多态性。方法: 采用PCR方法对汉族献血者CR1基因第26和29外显子进行扩增,并对扩增产物进行测序分析。结果: 180份汉族献血者中, Knops血型系统仅有York抗原(4223位)和KAM抗原(4870位)存在多态性,其中Yka+为48.89%,Yka-为51.11%;KAM+为79.17%,KAM-为20.83%;其余抗原均为纯合子。结论: 江苏汉族人群中的Knops血型系统仅有第4223位York抗原和4870位KCAM存在基因多态性。Abstract: Objective: The Knops blood group system consists of antigens encoded by exon 26 and 29 of complement receptor 1 (CR1) gene. To determine the frequency of seven single nucleotide polymorphisms (SNPs) in blood donors of Jiangsu and better elucidate the complexity of Knops group system. Method: Total 180 unrelated Han blood donors were recruited in Jiangsu. Specific primers in CR1 exon 26 and 29 were used genotyped Knops blood group system by polymerase chain reaction sequence-based typing. Amplification amplicons were purified and sequenced. Result: In Knops blood group system, the frequencies of the Yka+ and Yka- alleles were 48.89% and 51.11%; the frequencies of KAM+ and KAM- were 79.17% and 20.83%, other antigens were all hetozogotes. Conclusion: Seven SNPs of CR1 were identified in Jiangsu blood donors. The Knops blood group system in Jiangsu population had no diversity, except for KAM and York antigens.
