Significance of RhCE phenotype in identification for DEL among Han population in Hubei

摘要: 目的: 了解湖北汉族人群RhD与RhCE的分型特征,探讨其对Del的鉴定意义。方法: 采用抗人球蛋白卡式法对湖北汉族人群进行RhD初筛,IAT确认阴性标本,进一步做吸收放散试验确认Del,PCR法检测RhC(+)标本的RHD1227A等位基因。结果: 472例RhD阳性人群中共检出CCee 203例(43%)、CcEe 131例(27.7%)、ccEE 58例(12.3%)、Ccee 53例(11.2%)、ccEe 16例(3.4%)、CCEe 6例(1.3%)、CcEE 5例(1.1%);305例IAT确认阴性人群中共检出ccee 189例(61.9%)、Ccee 81例(26.6%)、ccEe 19例(6.2%)、CCee 10例(3.28%)、CcEe 6例(1.97%);63例经吸收放散试验确认Del组共检出Ccee 39例(61.9%)、CCEe 12例(19.1%)、CcEe 7例(11.1%)、CCee 5例(7.9%);Del组中RhC(+)比例为100%(63/63),RHD1227A阳性率为95.2%(60/63)。结论: 湖北汉族人群RhD抗原表达与RhCE分型有关;RhC(+)有助于Del型的筛选,可以结合吸收放散试验和检测RHD1227A来鉴定Del型。Abstract: Objective: To learn RhCE and RhD typing feature among Han population in Hubei and explore its significance for Del's identification. Method: RhD phenotype among patients was screened using anti-human globulin card method, negative specimens were confirmed by IAT, and Del was further identified with absorption elution test. RHD1227A allele of RhC (+) samples was detected by PCR assay. Result: A total of 472 cases of RhD-positive population were detected. Among them were CCee 203(43%), CcEe 131(27.7%), ccEE 58(12.3%), Ccee 53(11.2%), ccEe 16(3.4%), CCEe 6(1.3%) and CcEE 5(1.1%). 305 RhD-negative cases were confirmd by IAT, and among them were ccee 189(61.9%), Ccee 81(26.6%), ccEe 19(6.2%), CCee 10(3.28%) and CcEe 6(1.97%). Sixty-three Del cases were confirmed by absorption elution test,and among them were Ccee 39(61.9%), CCEe 12(19.1%), CcEe 7(11.1%) and CCee 5(7.9%). Del group RhC (+) ratio was 100%(63/63), and RHD1227A positive rate was 95.2%(60/63). Conclusion: RhD antigen expression was related with RhCE genotype among Han population in Hubei. RhC (+) would help to screen Del and can be combined with absorption elution test and RHD1227A to identify Del.
