摘要: 目的:对长链非编码RNA(lncRNA)8711-2进行生物信息学分析,预测其可能参与的生物学过程,并研究其在人脂肪组织细胞中的表达情况。方法:应用UCSC在线数据库获取lncRNA8711-2的序列、位置,并分析其特征;分离人脂肪组织,接种培养并诱导其向成熟脂肪组织细胞分化。采用逆转录PCR技术检测lncRNA8711-2在脂肪细胞分化中的表达变化。结果:lncRNA在脊椎动物中保守,与小鼠、大鼠、牛、恒河猴均具有较高的同源性,分析其序列所在的染色体区域,可能的调控靶标为Meox2基因。lncRNA8711-2在人脂肪前体细胞分化过程表达量明显增加。结论:lncRNA8711-2在人脂肪前体细胞分化过程表达量明显增加,预测lncRNA8711-2可能的调控靶标,参与人脂肪前体细胞诱导分化过程。

Abstract: Objective:To predict the biological process in which long non-coding RNA(lncRNA)8711-2 might involve by a series of bioinformatics analysis,and detect its expression in adipocytes,so as to lay foundations and provide theoretical basis for the further studies of lncRNA 8711-2 biological functions in preadipocytes and obesity.Method:The sequence,position,and structure characteristics of lncRNA 8711-2 were acquired from UCSC online databases,and target genes of lncRNA 8711-2 were predicted by UCSC databases.The adipose tissue was removed,inoculated and cultured,and induced to differentiate into mature fat cells from human scapula.The expression of lncRNA 8711-2 was detected using real-time quantitative polymerase chain reaction(PCR).Result:LncRNA had highly conserved property among different species,and the possible candidate target gene was Meox2 gene.The expression of lncRNA 8711-2 in adipocyte increased during the differentiation.Conclusion:The expression of lncRNA 8711-2 in adipocyte increased during the differentiation,which might play a crucial role in the process of preadipocyte-adipocyte transition via regulating the downstream target genes.-
