Frequency of killer cell immunoglobulin-like receptors and HLA-C in Jiangsu blood donors with HCV infection

  • 目的:自然杀伤细胞免疫球蛋白样受体(KIRs)是自然杀伤(NK)细胞上介导天然免疫的主要受体,同时KIRs也在T细胞上表达,与获得性免疫相关。本研究在对KIRs及其配体HLA-C进行分型的基础上,探讨KIRs、HLA-Cw的基因分型及其联合体与HCV感染的相关性。方法:采用序列特异性引物聚合酶链反应(PCR-SSP)法,对江苏献血人群中164例抗-HCV酶联免疫吸附试验(ELISA)双试剂反应性献血者(抗-HCV阳性组)和241例合格献血者(对照组)的KIRs和HLA-C的基因型进行检测和分析。结果:HLA-Cw的基因分型在抗-HCV阳性组和对照组中的分布无统计学意义;KIRs中与HLA-C配对的KIR基因分型仅有KIR2DS2在2组中的表达频率有差异(对照组21.2% vs抗-HCV阳性组37.8%),差异有统计学意义(P<0.01);HLA-C1C1与KIR2DS2的配对组合在2组中差异有统计学意义(22.1% vs 42.0%,P<0.05)。结论:江苏献血者人群中KIR2DS2可能是HCV感染的相关基因;HLAC1C1/KIR2DS2的配对组合可能激活了NK细胞和其他免疫细胞的功能,抑制病毒复制或促进HCV清除的功能,有待进一步实验证实。
