Effects of the Type IVB pili of Salmonella enterica serovar Typhi on the activity of PKC,ERK and NF-κB in THP-1 cells

摘要: 目的:初步探讨伤寒杆菌IVB型菌毛对THP-1细胞PKC、ERK、NF-κB活化的影响。方法:将THP-1细胞分别与有IVB型菌毛的A21-6菌、缺失IVB型菌毛的pilS-菌共同孵育,底物磷酸激酶测定法检测PKC活性变化,免疫印迹法检测ERK磷酸化水平,荧光素酶报道基因分析法检测NF-κB激活表达。结果:THP-1细胞经有IVB型菌毛的A21-6菌刺激后,PKC活性变化、ERK磷酸化水平和NF-κB激活表达均显著高于相应的处理组;PKC、ERK、NF-κB活化水平分别于刺激10、30、60 min后即明显升高;并分别于30、60、120 min左右达到峰值。结论:伤寒杆菌IVB型菌毛显著增强了THP-1细胞PKC、ERK、NF-κB活化水平,IVB型菌毛在伤寒杆菌诱导单核/巨噬细胞PKC、ERK、NF-κB活化过程中是一种重要的强刺激因素。

Abstract: Objective:To investigate the effects of the Type IVB pili of Salmonella enterica serovar Typhi on the activity of PKC, ERK and NF-κB in THP-1 cells.Method:THP-1 cells were incubated with the Salmonella Typhi strain A21-6 or Salmonella Typhi strain pilS-,the activity of PKC, ERK and NF-κB were measured with substrate phosphorolysis kinase assay,Western blotting and luciferase reporter gene assay,separately . Result: The Type IVB pili of Salmonella entrica serovar Typhi strain A21-6 was significantly effected on the activity of PKC, ERK and NF-κB;the activity of PKC, ERK and NF-κB were increased 10min、30min、60min after being stimulated with the Salmonella Typhi A21-6,and reached the peak value at 30min, 60min and 120min,respectively. Conclusion: The type IVB pili of Salmonella Typhi may play an important role in the activity of PKC, ERK and NF-κB induced by Salmonella Typhi,which could significantly increase the activity of PKC, ERK and NF-κB in THP-1 cells.-
